Hematologic toxicity of oxoplatina in experiment by chronical administration

Summary: It was examined a haematological toxicity of oxoplatin during repeated administration to mice in the dosis LD10 in regimen qd x 5. The time of the experiment was 30 days.
Our results showed a significant decrease of erythrocytes, haemoglobin concentration and hematocrite in period from the 3rd to 19th day in comparison to values of controls. The number of reticulocytes decreased until the fifths day than from the 7th day an increase was observed. The thrombocytes decreased on the 10th day only. The decrease of leucocytes was observed from the first day, until the end of the experiment return back to reference values. In the diíferential count of peripheral blood, the decrease of segmente forms of leucocytes appeared with the maximum in the fifth day and was accompanied by polychromasia, anisocytosis and increased number of Howell - Jolly bodies. In the femur bone, the significant decrase of all of absolute number of nucleated blood cells in the first 5 days was observed, with the decreased mitotic activity in comparison to controls. The decrease in the red blood cells was more pronounced. From the 7 day the differences in the findings from the femur bone disappeared.
