Identifi cation of individual myeloma-specifi c T cell clones by molecular analysis of the T cell receptor beta (TCRB) is the most sensitive method for the anti-tumor immune response monitoring not only in multiple myeloma. This novel approach called clonotypic assay is based on the fact, that the TCRB sequence is unique for every T lymphocyte clone and thus can serve as an ideal candidate for
the individual T cell clone monitoring. As an example, the immune response against myeloma antigens was tested and demonstrated an oligoclonal myeloma-specifi c response, ie. only a few immunodominant myeloma-specifi c T cell clones were activated. This fi nding corresponds with the assumption of limited antigenicity of myeloma cells. A detailed study of anti-tumor immune response
based on exact myeloma-specifi c T lymphocytes idenfi cation and their in vivo monitoring can be useful tool for precise and highly sensitive immunomonitoring in adoptive immunotherapy protocols recently used in multiple myeloma.
